Written by Suleman Shahid
Tuesday, 14 February 2006

Job description
• Swammerdam Institute for Life Sciences (SILS)
• Department of Plant Physiology
• full-time (38 hours per week) for a period of 18 months

The aim of the project is to identify transcription factors involved in the regulation of flavour production in tomato fruit. The technician will operate within the Centre of Biosystem Genomics (CBSG), in the tomato quality project – a collaboration between the UvA, Plant Research International (Wageningen) and Keygene (Wageningen). The project aims to clone fruit-specific transcription factors from tomato cultivars with contrasting flavours. Homology-based PCR experiments and microarray experiments will provide a comprehensive set of candidate transcription factors that are fruit-specific and differentially expressed. The collection of cDNAs will be 1) overexpressed using fruit-specific and inducible promoters in tomato plants, and 2) used for RNAi constructs. Technical approaches include PCR, gene cloning, microarray hybridizations, vector construction, transformation of tomato cultivars, analyses of transgenic plants.
