Netherlands: Postdoc in neurophysiology

It is now firmly established that
activity-dependent modulation of presynaptic secretion is a central
aspect for real-time information processing in the brain. However, the
intracellular signal transduction cascades responsible for this
modulation are poorly understood. We have recently characterized some
of the genes required for Ca2+- and phospholipase C-dependent
modulation (Wierda et al. 2007, Neuron 54 pp275). This project aims to
identify the cross-talk between these genes and other protein kinase
pathways in the terminal and their regulation by presynaptic receptors
and proposed retrograde signaling.
Description of work
This project uses patch clamp and null mutant rescue approaches in
microdot island cultures (autapses). Several presynaptic genes have
been silenced and viral based rescue construct are available. Some of
the experiments will also combine synaptophysin-pHluorin based imaging
of secretion. Autaptic neurons and viral stocks are in principle
supplied by technicians.
All the patch clamp and optical equipment and analysis software are available from the start of the project.
Requirements
University Graduate We are looking for
candidates with a PhD in neuroscience or physiology, extensive
experience with patch clamp and a strong motivation to pursue a career
in science. Some experience with cell culture and life cell imaging are
beneficial.
